Five placebo-controlled phase 2 studies were undertaken in which efficacy and safety of Cholestagel was evaluated. The Phase 2 studies are regarding objectives, treatment, study design and endpoint assessment similar to the two pivotal Phase 3 studies. For this reason all Phase 2 and 3 studies are presented and discussed combined in the following sections. Main studies Phase 2 and Phase 3 ; Description of the studies Study GTC-48-301 was a pivotal Phase 3 dose-response study designed to determine the long-term efficacy and safety of Cholestagel. Study GTC-48-302 was a pivotal Phase 3 dose-regime study, designed in a similar manner as GTC-37-202, to confirm the efficacy of once a day and split daily doses. Three studies with colesevelam and HMG-CoA reductase inhibitors GTC-37-203, GTC-48204, GTC-48-205 ; are also considered as main studies, as they have been conducted to support the add-on indication. Two open-label studies GTC-37-901 and GTC-44-201 ; are considered as supportive efficacy studies. All studies, except GTC-37-901 and GTC-44-201, were double blind and placebo-controlled. In some studies, patients were stratified prior to randomisation into categories of baseline LDL cholesterol. No comparative studies with other bile acid sequestrants were performed. In all studies, all patients underwent a physical exam before and after therapy and a medical history was obtained. All patients were placed on a controlled diet low in fat and dietary cholesterol for at least 2 weeks prior to dose initiation and all patients were required to adhere to a low-fat, lowcholesterol diet while being enrolled in the studies. At least two baseline lipid values were obtained, and serum lipid values during treatment were obtained at intervals of at least 2 weeks duration.

Furthermore, computer-based image processing software, with the capture program to choose the capture interval and period, and analysis techniques were developed for quantitative analysis of the cell migration on extracellular matrices. The results showed that the migration speed of L-929 cells on the collagen-coated glass was significantly p 0.05 ; increased compared to the non-coated control. On the morphological observations, it was showed that the cells on the collagen-coated glass looked much healthier than those on the control. The investigation will be done in the frame of a PhD thesis jointly carried out by PSI LAC and EPFL LPAS. EMPA uses the same dispersion model in time-inverted mode in the frame of source apportionment studies. All four institutions will benefit from an improved version of the current turbulence schemes. After having become familiar with the main features of the meteorological and photochemical dispersion models used at PSI, the PhD student will spend a few weeks at EPFL to become acquainted with turbulent transport and its parameterization. He will implement the most promising schemes into an interface that links MM5 and CAMx. In the first phase the various approaches shall be tested for a given domain including complex topography, urban and rural areas. Their impact on meteorology and finally on the concentrations will be investigated and the optimum parameterization will be determined. In a second phase the model package shall be applied to more extended domains covering Switzerland and northern Italy.
